Description GAP-43 or growth associated protein 43 (also referred to as neuromodulin, B-50, p57 and F1) is a nervous system-specific phosphoprotein and a major component of motile growth cones which form the leading edge of the elongating axon. GAP-43 is closely associated with the membrane skeleton and has been implicated in regulating interactions between cytoskeletal components and the plasma membrane. Interestingly, axonal transport of GAP-43 occurs only in neurons that are extending axons during development or regeneration after injury. Extracellular signals modify the activity of the GAP-43 protein in several ways, which include phosphorylation, calmodulin (CaM) binding, and Ca2+-dependent proteolysis. For example, GAP-43 is phosphorylated on Ser41 in response to activation of protein kinase C (PKC). Phosphorylation of GAP-43 by PKC is restricted to the distal axon and growth cone and can be stimulated both by targetderived chemotropic factors, as well as by neuronal cell adhesion molecules. Ser41 phosphorylated GAP-43 is predominately localized to areas where the growth cone makes stable interactions with the substratum or with other cells. In contrast, un-phosphorylated GAP-43 is localized to areas of the growth cone that are retracting. Phosphorylation of GAP-43 on Ser41 also functions as a switch to inhibit GAP-43 from binding to CaM. Unphosphorylated GAP-43 interacts with CaM through an IQ motif and has a higher affinity for CaM in the absence rather than the presence of calcium.Immunogen: Antibody was raised against purified full length GAP43 protein (both phosphorylated & unphosphorylated forms).
