Format Purity: Greater than 98% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Format: The protein was lyophilized from a concentrated (1 mg/ml) solution with no additives.
Protein Family: Belongs to the IL-6 superfamily.

Protein Content: Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.95 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC. using a calibrated solution of IL-11 as a Reference Standard.

Endotoxin: Less than 0.1 ng/ug (IEU/ug) of rHuIL-11.

Molecule Name Interleukin-11 (IL-11)
Source/Expression System E. coli
Unit 0.002 mg
Description Function: Directly stimulates the proliferation of hematopoietic stem cells and megakaryocyte progenitor cells and induces megakaryocyte maturation resulting in increased platelet production.Subcellular Location: Secreted protein.Pharmaceutical: Available under the name Neumega (Wyeth). Used for the prevention of severe thrombocytopenia and the reduction of the need for platelet transfusion following myelosuppressive chemotherapy.Similarity: Belongs to the IL-6 superfamily.Summary: The protein encoded by this gene is a member of the gp130 family of cytokines. These cytokines drive the assembly of multisubunit receptor complexes. all of which contain at least one molecule of the transmembrane signaling receptor IL6ST (gp130). This cytokine is shown to stimulate the T-cell-dependent development of immunoglobulin-producing B cells. It is also found to support the proliferation of hematopoietic stem cells and megakaryocyte progenitor cells. Chromosome Location: 19q13.3-q13.4 Sequence Length (aa): 199 MW: 21429KEGG pathway: Cytokine-cytokine receptor interaction 04060KEGG pathway: Hematopoietic cell lineage 04640KEGG pathway: Jak-STAT signaling pathway 04630 OMIM: 147681; gene. [ NCBI / EBI]human Storage: 4°C for short term (weeks) and -20°C for long term. Avoid frequent freeze and thaw. Stability: 5 years at -20°C. Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Gly-Pro-Pro-Pro-Gly. N-terminal methionine has been completely removed enzymatically.Biological Activity: rHuIL-11 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of the proliferation of murine 7TD1 was found to be less then 10ng/ml. corresponding to a Specific Activity of 1 x 10 IU/mg.Solubility: It is recommended to reconstitute the lyophilized rHuIL-11 in sterile 18MolarOmega-cm H2O not less than 100ug/ml. which can then be further diluted to other aqueous solutions.Aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
